Effects of Physalis Angulata Extract on Pulmonary Fibrosis in a Scleroderma Rat Model

Abstrak
Introduction: Scleroderma is an immune-mediated connective tissue disease with interstitial lung disease (ILD) being its leading cause of mortality. However, the treatment of ILD in scleroderma is still unsatisfactory. Physalis angulata (ciplukan) herb has shown promise in treating skin sclerosis in scleroderma patients but its effect on lung fibrosis remain unexplored. The aim of this study is to evaluate the effect of ciplukan herb for the treatment of lung fibrosis in bleomycin-induced scleroderma animal model. Methods: Sprague-Dawley rats were divided into 6 groups: 4 groups were given bleomycin and ciplukan extract dose 50,100,150, and 200 mg/kg Body Weight, respectively, bleomycin only and negative control. Bleomycin was given subcutaneously for 14 days and ciplukan or saline were given from day 21 for 30 days and then animals were sacrificed. Degree of fibrosis and area of fibrosis from lung histopathology, TGF-β, and MMP-13 levels from lung homogenates were then analyzed. TGF-β and MMP13 were measured by ELISA methods. Results: There was significant reduction in lung fibrosis extent on groups receiving ciplukan extract dose 100, 150, and 200 mg/kgBW compared with bleomycin group however dose 200 mg/kgBW showed increase in fibrosis score. There were no differences of TGF-β and MMP-13 levels among groups. Conclusion: Ciplukan extract reduced extent of lung fibrosis but showed no effect on TGF-β and MMP-13 levels. © 2025 Universiti Putra Malaysia Press. All rights reserved.
